Looking at the file /opt/local/Library/Frameworks/Python.framework/Versions/2.7/share/pygobject/2.0/defs /gio-types.defs It is clear that this patch is no longer being applied in more recent ports e.g. grep define-enum /opt/local/Library/Frameworks/Python.framework/Versions/2.7/share/pygobject/2.0/defs /gio-types.defs | grep -i flag (define-enum MountMountFlags (define-enum DriveStartFlags (define-enum SocketMsgFlags which is precisely what get's replaced by the previous patch. I haven't checked to see if the old patch has the right line numbers but it should be easy enough to fix. I also haven't checked earlier python versions of gobject.
