Ok, it's having trouble running the reinplace command in the port's post-destroot procedure.

Not weird; when you build the code by hand, you're not running the reinplace command that's in the portfile.

Not necessarily. The port may be broken in same cases. Does the file "/opt/local/var/macports/build/_opt_local_var_macports_sources_rsync.macports.org_release_ports_databases_couchdb/work/destroot//opt/local/Library/LaunchDaemons/org.apache.couchdb.plist" exist? If not, what's the longest part of that path that does exist?
